CD9 cross-linking through N- or C-terminal cysteine residues. RD cells were transfected with constructs encoding wild-type CD9 protein, CD9 containing only the three N-terminal cysteines (Cys9, 78 and 79), or the three C-terminal cysteines (Cys87, 218 and 219). Cells were treated with 2-BP for 16 hours and lysed in buffer containing 1% Brij97 for spontaneous disulfide cross-linking. Cell lysates were analyzed by CD9 Western. Relative intensity of bands corresponding to CD9 monomers, dimers, trimers and tetramers was calculated as a percent fraction of total protein material (monomers through tetramers).
