GdPDC characterization data. A) The temperature (■) and pH (♦) profiles of GdPDC using pyruvate as substrate. The assay is described in the methods section. 100% activity is analogous to a specific activity of 60 U/mg for Topt and 36 U/mg for pHopt. B) Thermal inactivation GdPDC at 25°C (♦), 40°C (■), 55°C (▲) and 60°C (×). Enzyme activity at zero time using the standard assay at 25°C is set to 100%, analogous to a specific activity of 10 U/mg. Assays were performed in 200 mM citrate buffer at pH 6.0 and pyruvate as substrate. The data represents the average of at least three individual rounds of protein purification and assay.
